Fluorimetric determination of amino acids and photochemical stability of the products of their reaction with orto-phthalic aldehyde under irradiation of a high-power pulsed laser.

Abstract

The possibility of high-sensitive determination of amino acids was investigated with the use of fluorescence of their derivatives obtained by reaction of aqueous solution of ortho-phthalic dialdehyde and nucleophilic additives: potassium cyanide, sodium sulfite and N-acetyl-L-cysteine. With glycine as an example, it was shown that chemical stability of yielded highly fluorescent compounds (substituted isoindoles) and their photochemical stability under irradiation of a powerful pulsed laser is substantially higher than the ones of widely used reagent for determination of amino acids - the basic aqueous solution of ortho-phthalic dialdehyde and 2-mercaptoethanol. The analytical characteristics of the newly developed reagents are the same or better than of the standard one.
